Abstract

Humanized mice, distinguished by the engraftment of functional human cells and tissues into immunodeficient mouse hosts, represent a paradigm shift in preclinical animal models. This paper offers a comprehensive examination of the development and multifaceted applications of humanized mice, emphasizing their pivotal role in advancing our understanding of infectious diseases, cancer, regenerative medicine, graft-versus-host disease, allergies, and immunity. Originating from the introduction of immunodeficient mice bearing mutations in the IL2 receptor common gamma chain (IL2rgnull) in the early 2000s, humanized mice have evolved into versatile platforms for disease modeling, contributing significantly to both basic and translational research.
